Abstract

The main objective of this study was to determine the characterization of ketoprofen – hydroxypropyl-β-cyclodextrin inclusion complex. Solvent evaporation method was chosen because it is quite simple and economic both on laboratory and large scale production and is considered alternative to the spray drying technique. Solubility studies demonstrated the formation of the ketoprofren – hydroxypropyl-β-cyclodextrin inclusion complex with 1:1 stoichiometry. Equimolar ketoprofren – hydroxypropyl-β-cyclodextrin were prepared using solvent evaporation method and then characterized by X-ray Diffraction, Differential Thermal Analysis (DTA), and Infrared Spectrophotometry compared with ketoprofen, hydroxypropyl-β-cyclodextrin, and the physical mixture of ketoprofren and hydroxypropyl-β-cyclodextrin. The result of the inclusion complex characterization performed with X-ray diffractometer showed significant decrease of ketoprofen’s cristallinity degree compared with ketoprofen, but the physical mixture showed the superimposition between ketoprofen and hydroxypropyl-β- cyclodextrin’s diffractogram. Analysis using differential thermal analysis method showed one new endothermic peak at 130.7oC while in the physical mixture had two peaks at 82.6oC and 132oC. The characterization using infra-red spectrophotometer showed that the specific band of ketoprofen disappeared in the inclusion complex’s spectra while the physical mixture’s spectra showed the superimposition between ketoprofen and hydroxyprophyl-β-cyclodextrin’s spectra. All methods showed that the inclusion complex has been formed.
