Abstract

Inflammatory pain is defined as unpleasant experience due to tissue damage caused by inflammation. Pathophysiology of pain caused by an activation of NMDA receptor followed by increase of Ca2+ influx which may increase pain intensity and in the long term will lead to chronic pain that is accompanied by synaptogenesis process. The effect of music on decreasing pain intensity was investigated. Therefore, this study was conducted to determine the effect of frequency and amplitude of music to decrease pain intensity and synaptogenesis process. Model of inflammation was induced by intraplantar injection of Complete Freund’s Adjuvant (CFA) on the hind paw of mice. Mice were divided into 5 groups : control group, CFA group, CFA with rock, classical and javanese music treated-group. Music was given one hour daily at day 7 to 13 after induction CFA. Inflammation characterized by increased in paw thickness and decreased in mice’s latency on thermall stimulation. The result showed no significant difference in mice’s latency on thermal stimulation between CFA group and CFA with rock music treatedgroup. Whereas on CFA with classical music treated-group (p <0.001) and CFA with javanese music treated-group (p <0.01) showed significant differences. As for the decreased in hind paw thickness did not show significant difference (p> 0.05) in all groups. Inflammation also leads to the morphological changes in spinal cord and alters the number of inflammattory cells. Regarding immunohistochemical observations, CFA with classical music treated-group (p <0.01) showed significant decreased in synaptogenesis process. Whereas in CFA with gamelan music treatedgroup and CFA with rock music treated-group did not show significant decreased. It is proved that the difference of frequency and amplitude of music gives an effect on pain conditions through the difference of molecular mechanisms.
