Abstract

Salysilic acid as a drug compound has been developed but not showed the optimal result. The purpose of this research is to synthesize 5-nitro-2-(octanoyloxy) benzoic acid as salysilic acid derivate and examine its analgesic activity in mice. 5-nitro-2-(octanoyloxy) benzoic acid was synthesized with esterification method by reacting 5-nitro salicylic acid and octanoyl chloride. Pyridine and tetrahydrofuran was used as respective catalyst and solvent. The result obtained for the synthesize of 72%, in the form of white powder. The resulted compound was recrystallized by using ethanol and its purity was tested by thin layer chromatography and melting point test. The structure was confirmed by UV spectrophotometry method, infrared spectrophotometry method, and 1H-NMR spectrometry method. Its analgesic activity was tested with Writhing test method. The pain-inhibition percentage of 5-nitro-2-(octanoyloxy) benzoic acid for 25mg/kg body-weight was 47.62%. In the dose 50 mg/kg body-weight was 76.43% and for 100 mg/kg body-weight was 88.33%. In other hands, the standard compound acetosal in the dose 25mg/kg, the pain-inhibition was 32.74%, 50 mg/kg body-weight was 60.71% and 100 mg/kg body-weight was 77.02%. ED50 of 5-nitro-2-(octanoyloxy) benzoic acid is 24,62 mg/kg lower than 5-nitro salicylic acid 40,38 mg/kg. It concluded that 5-nitro-2-(octanoyloxy) benzoic acid have higher analgesic activity than acetosal.
