Abstract

In the world, breast cancer in women is the most potentially happened than other cancers with one survival of six diagnosed patients in recent five years. It was found that breast cancer cells is excess with folic acid receptor, neither on the others cancer cells nor normal cells. So it is necessary to modify the folic acid molecule to produce another analog that is expected to provide a spesific and optimal activity. Folic acid modification begins with drafting a compound where folic acid is added the groups which have lipophilic, electronic and steric factors. Tested in silico to predict bond strength analogous to receptors. The result of rerank score of 10-N-(4’chlorobenzoyl)folic acid has lower than methotrexate. It can be predicted that 10-N-(4’chlorobenzoyl)folic acid has higher activity than methotrexate. The synthesis of these compounds used acylation method. Results compound of synthesis is 10-N-(4’chlorobenzoyl)folic acid with 14% yield and proved by TLC and 1H-NMR spectrophotometric test. The compounds tested with in vitro assays using MTT assay test against MCF 7 cells that compared with methotrexate can not be analyzed because the method does not show the good inhibition against 10-N-(4’- chlorobenzoyl) folic acid and methotrexate.
