Abstract

Background: Stroke is one of the common causes of disability and death in worldwide. Ischemic stroke comprises more than 70% of all type of stroke. The present study was designed to investigate the effect of erythropoietin (EPO) on cognitive function, infarct lesion, histopathological changes, and the level of α-melanocyte-stimulating hormone (α-MSH) in plasma after stroke induced by right unilateral common carotid artery occlusion (RUCCAO) method. Materials and Methods: Male balb/c mice were divided into five groups (n=6)i.e. sham, stroke, stroke with EPO 2,000, 5,000, and 10,000 IU/kg groups. The stroke model was performed by blocking the right common carotid artery for 2 hours. EPO-treated groups received a single intraperitoneal injection of recombinant human EPO one hour after surgery. Cognitive impairment by Ymaze plus cues method and level of α-MSH were evaluated on days 1, 3, and 7. Infarct area and histological change were measured on day-7 after stroke induction. Results: In this study, the treatment of EPO has shown an improvement in cognitive function on days 1, 3, and 7. Furthermore, EPO reduced infarct lesion and the neuronal damage in the CA1 of the hippocampus. Moreover, the plasma level of α-MSH on the EPO treatment groups was significantly higher than the stroke group on day-7 after stroke induction. Conclusion: EPO may have a neuroprotective effect on ischemic stroke condition, possibly through the α-MSH upregulation.
