Abstract

The aim of the study was to determine the effect of different polymers, different consentrations of polymer, the size of beads and duration of milling on the particle size reduction of mefenamic acid. Stability of mefenamic acid nanosuspension during storage as well as the influencing factors were also investigated. Mefenamic acid was milled using stirred beads in a polymer solution. Differential Thermal Analysis were used for qualitative assay of mefenamic acid whilst Photon Correlation Spectroscopy and light microscope were used to evaluate particle size after predetermined time of milling and storage. Stability of mefenamic acid nanosuspension was evaluated for 14 days storage at room temperature and at 8ºC. Minitab statistical software 19.0 were used to analyze which factor that has a significant effect on particle size reduction. In total, there were 8 formulas evaluated. Based on pareto chart, different size of milling beads has a significant impact in particle size reduction whilst polymer type and concentration influenced particle size stability during storage. Interestingly, mefenamic acid nanosuspension stabilized with PVP K-30 showed particle size shrinkage during storage. In general, particle size of mefenamic acid nanosuspension at 8°C was more stable. Based on the contour plot analysis, smallest particle size of mefenamic acid nanosuspenson (<350 nm) could be obtained by using 10% w/w Kollicoat IR as stabilizing agent and milled for 6 hours using beads milling size 0.5 mm.
