Abstract
The aim of this study was to determine the profile of the Luteinizing Hormone Receptor (LHR) gene in Madrasin cross breed cattle. The sample of this study used blood samples from 3 breeds of cattle. There were Madrasin cattle from Desa Galis, Kabupaten Pamekasan, Madura cattle from UPT Pembibitan dan Kesehatan Hewan, Kabupaten Pamekasan and Limousin cattle from BET Cipelang Bogor. The sample criteria were used female cattle around 2,5 – 3 years old and ever partus (once). The genomic DNA was extracted for polymerase chain reaction (PCR), followed by sequencing with software BioEdit Vers. 8.0 to determine the sequence of nucleotide bases. The results of this study were that three samples detected fragments in the form of DNA with nucleotide base of 373 bp and the amino acid length is 125bp. Based on the phylogenetic tree, the LHR gene in Madrasin cattle is in one branch with the Limousin cattle LHR gene. In conclusion, the LHR gene in Madrasin cattle can be detected using the PCR method which nucleotide base has length of 373bp and amino acid 125bp. The LHR gene does not have M (Methionine) amino acid. There were change variable site of amino acid at position 22 Serine (S), position 46 Threonin (T) and position position 85 Cystein (C). The LHR gene in Madrasin cattle has a closer relationship (phylogenetic) with Limousin cattle and the homology value is 77%.
