Abstract

Quercetin or 2-(3,4-dihydroxyphenyl)-3,5,7-trihydroxy-4H-chromen-4-one is a compound that has been commonly utilized in medical field due its various functions. Quercetin is classified as class II BCS which has low solubility but good permeability. Solid dispersion is successfully applied to improve the solubility and consequently the bioavailability of poorly water soluble drugs. The purpose of this research was to increase the solubility and dissolution rate of quercetin by solid dispersion system of quercetin – HPMC 3 cps with different ratios of quercetin – HPMC 3 cps : 1:1, 1:2 and 1:3 (w/w). The solid dispersion system was prepared by solvent evaporation method then solubility and dissolution test were performed. The solubility medium was 40 mL of citric acid – NaOH (pH 5,0 ± 0,05), maintained at 30 ± 0,5 oC for 240 minutes. The dissolution medium was 900 mL of 1 % SLS in water, maintained at 37 ± 0,5 oC for 30 minutes and the stirring speed was 100 rpm. For comparison purpose the solubility and dissolution test was also performed on : solid dispersion of quercetin – HPMC 3 cps, physical mixture and pure quercetin. The results showed that the solubility of solid dispersion quercetin – HPMC 3 cps (1:3) increased 3,50 times higher than pure quercetin. For dissolution rate, solid dispersion quercetin – HPMC 3 cps (1:3) increased 1,42 times higher than pure quercetin.
