Abstract

Ezetimibe is the first line lipid-lowering drug that inhibits the intestinal uptake of cholesterol without affecting the absorption of fatsoluble nutrients. Ezetimibe being practically insoluble in water, exhibits a low dissolution profile. Solid dispersion system could enhance dissolution rate of ezetimibe. A combination of melting and adsorption techniques increases the effective surface area of ezetimibe leading to improved dissolution. The aim of this investigation was to improve the dissolution rate of ezetimibe by preparing solid dispersion with poloxamer 188 as the matrix and absorbed solid dispersion using lactose monohydrate and Avicel PH 101 (2:1) as the adsorbent. Dissolution of pure ezetimibe, solid dispersion and adsorbed solid dispersion were evaluated. The physical characterisation by powder X-ray diffraction (PXRD) and differential thermal analysis (DTA) were evaluated. The result showed that the dissolution rate of ezetimibe in adsorbed solid dispersion was significanlty higher than the pure ezetimibe and ezetimibe-poloxamer 188 solid dispersions.
