Abstract

Subchronic toxicity test of diterpene lactone fraction of Andrographis paniculata Nees. was done to know the subchronic toxicity effect on rat with the parameter of toxicity including clinical chemistry (SGOT, SGPT, BUN, Creatinine, and protein total), hematology (hemoglobin, platelets, and leukocyte), and histopathology of liver and kidney. subchronic toxicity test was done for 30days in three group of rat. each group consist of 5 male and 5 female rats. the test samples was given orally in CMC Na 0,5% suspension, with the appropriate dose in every groups. one froup is control which was given CMC Na 0,5% suspension without diterpene lactone fraction of Andrographis paniculata Nees. two other groups were given CMC Na 0,5% suspension with diterpene lacton fraction of Andrographis paniculata Nees. which equivalent to 500 mg/kgBB and 1000 mg/kgBB rat. the rats were weighed every week for adjustment of dose. at the end of treatment, all rats were sacrified by anasthesia with ether. the examination of clinical chemistry parameters (SGOT, SGPT, BUN, Creatinine, and Protein total), hematology parameters (hemoglobine, trombocyte, and leukocyte), and histopathology of liver and kidney from the rat was done. data was analyzed with 95% degree of One-Way ANOVA, Independent-Samples T Test, and Kruskal Wallis. From result, the difference between the test and control groups were statically not significant. Except on SGPT, Protein total, Leukosit, and Monosit which significant lower than other groups. But that level is still on physiological range. So that it can be concluded that diterpen lacton fraction of Andrographis paniculata which equivalent to 500 mg/kgBB and 1000 mg/kgBB didn’t caused subchronic toxicity on rat with SGOT, BUN, Creatinine, Hemoglobin, Trombocyte, Eusinofil, Neutrofil, and histopathology of liver and kidney as the parameters of subchronic toxicity.
