Abstract

The aim of this research was to prove the effect of Roselle calyx (Hibiscus sabdariffa L.) extract against Salmonella pulorum in vitro. The method consisted of two steps, first antibacterial determination of Roselle calyx extract by diffusion disc with 100% concentration was used Chloramphenicol 30 μg as positive control and the second step determined of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of Roselle calyx extract by dilution against Salmonella pullorum. The concentration of Roselle calyx extract for determination of MIC and MBC were 6.25%, 12.5%, 25%, 50%, 100% with 4 repetition and Salmonella pullorum suspension as negative control. The result showed that Roselle calyx flower red (Hibiscus sabdariffa L.) extract could inhibited and killed the growth of Salmonella pullorum by diffusion and dilution method. The clear zone diameter of Roselle calyx extract with 100% concentration were 26mm, 24mm and 26mm while Chloramphenicol as positive control showed resistance to bacteria. The result of dilution method showed that Roselle calyx extract has antibacterial activities against Salmonella pullorum with the Minimum Bactericidal Concentration (MBC) is 50%.
