Abstract

Epidermal Growth Factor is polypeptide that act both as paracrine and autocrine factor in regulation of cell growth and oocyte maturation. The aim of this research was to identify a protein 46 kDa that estimated as Epidermal Growth Factor isolated from in vitro maturated bovine oocyte with electrophoresis method. Bovine ovary (from a slaughterhouse) was aspired in its ovary follicle ≤ 5 mm with spuit and needle. The oocyte were matured in Tissue Culture Medium 199 for 22 hours at 38.50C in incubator CO2. The protein 46 kDa was prepared by using Sodium Dodecyl Sulphat Polyacrylamide Gel Electrophoresis (SDS PAGE). Based on the calculation of regression equation from protein marker, 12 fractions were determined, 172.7 kDa, 153.09 kDa, 118.24 kDa, 102.43 kDa, 89.27 kDa, 59.75 kDa, 46.41 kDa, 43.82 kDa,40.2 kDa, 36.06 kDa, 23.45 kDa and 18.42 kDa. The molecular weight of 46.41 kDa that appeared in the protein band was identified as protein that estimated EGF playing role in maturation process.
