Abstract

The aim of this research was to investigate the effect of various concentrations using ethylene glycol (EG) on vitrification to viability of mice embryo post thawing. The collected embryos were washed using PBS and divided in three groups which are EG 10%, 20% and 30%. Embryos were exposed inside cryoprotectant at 200C for 5 minutes. It were putted inside the straw than added by 0.5 M sucrose. Straws were exposed 10 second, then directly inside liquid nitrogen. Frozen embryos were thawed in waterbath at 370C for 30 seconds. It were washed sequently by 0.5 M and 0.1 M of sucrose to removed cryoprotectant. Inverted microscope were used to examine the viability of mice embryo. The result showed that there was significantly different (p < 0.05) between various concentration using EG. Rate of viability were 69.22+6.88, 90.00+8.94, 77.77+9.93. Therefore, there was effect from various concentration EG on vitrivication to viability of mice embryo post thawing, with the highest percentages of viability were 20%.
