Abstract

This research aim is to produce polyclonal antibody of whole protein of adult Haemonchus contortus. The polyclonal antibody is applicable to haemonchosis serologic diagnosis development at goat and sheep. Whole protein was isolated from adult H. contortus which taken from the abomasum of goat and sheep which taken on Surabaya’s Slaughter House. Protein extraction was made by sonication technique. Protein fractination was performed by SDS-PAGE. Polyclonal antibody production started at immunized rabbit with 100 μg dose of H. contortus whole protein added with complete adjuvant. Booster conducted one week after first injection with same dose of H. contortus whole protein added with incomplete adjuvant. Booster repeated one week after previous until 9th booster. Titer of antibody was examind by Indirect Enzyme Linked mmunosorbant Assay (ELISA) method. Result of ELISA after the 9th booster showed the increase of antibody titer compared with the control serum.
