Abstract
Partial purification of xylanase enzyme from Pacet Hot Spring, East Java have been done in this research. The purpose of this research is knowing purification level of xylanase enzyme which isolated from Pacet hot spring, East Java by hydrophobic interaction chromatography and ion exchange chromatography method. The steps of partial purification was carried out by addition of ammonium sulfhate and followed by dialysis, interaction hydrophobic chromatography and anion exchange chromatography. The result shown that there was an increasing purity level of xylanase enzyme from crude extract as much as 62,18 after hidrorobic interaction hidrofobic chromatography has been done. The result of chromatography hidrofobic shown there was two activity of xvlanase enzyme in 2nd and 6th fraction. Eventhough anion exchange chromatography need optimation to get xylanase enzyme fraction.
