Abstract

Penelitian ini bertujuan untuk mengetahui kemampuan enzim xilanolitik rekombinan dari E coli DH5 a (pTP510) dalam menghidrolisis beberapa jenis xilan komersial dan menganalisis produk gula pereduksi yang dihasilkan. Enzim xilanolitik rekombinan (β-xilosidase dan α-L-arabinofuranosidase) mampu menghidrolisis beberapa jenis xilan komersial (oat-spelt xylan, birchwood, wheat, rye, dan arabinan) dengan aktivitas xilanolitik yaitu : oat-spelt xylan (173,33 U/mL), birchwood (92,22 U/mL), wheat (652,22 U/mL), rye (494,44 U/mL), dan arabinan (340 U/mL). Uji aktivitas enzim xilanolitik menggunakan substrat spesifik p-nitrofenil-β-D-xilopiranosida (pNP-x) menunjukkan aktivitas xilosidase sebesar 1,5869 μ mol/menit (Unit). Produk hidrolisis dianalisis dengan HPLC. Hasil menunjukkan bahwa xilosa, arabinosa, dan xilo-oligosakarida dihasilkan dari hidrolisis birchwood, wheat, rye, dan arabinan, sedangkan xilosa dan arabinosa dihasilkan dari hidrolisis oat-spelt xylan. Translation: The purposes from this research were to know the ability of recombinant xylanolytic enzyme from recombinant E.coli DH5α (pTP510) to hydrolyze several commercial xylan and analysis the reduction sugar product.Recombinant xylanolytic enzyme (,β -xylosidase and α -L-arabinofuranosidase) could hydrolyzed several commercial xylan (oat-spelt xylan, birchwood, wheat. rye, and arabinan) with xylanolytic activities are : oat-spelt xylan (173,33 U/mL), birchwood (92,22 U/mL), wheat (652,22 U/mL), rye (494,44 U/mL), and arabinan (340 U/mL). Xylanolytic enzyme assay use specific substrats p ¬nitrofenil-β-D-xilopiranosida (pNP-x) shown xylosidase activity 1,5869 μ mol/menit (Unit). Hydrolysis product was analyzed by HPLC. The result showed xylose, arabinose, and xylo-oligosaccharide was produced from birchwood, wheat, rye, and arabinan, although xylose and arabinose produced from oat-spelt xylan.
