Abstract

Biosurfaktan Pseudomonas aeruginosa IA7d memiliki kemampuan untuk menghambat pertumbuhan mikroba. Penelitian ini bertujuan untuk mengetahui potensi penghambatan dan nilai MIC (Minimal Inhibitory Concentration) biosurfaktan Pseudomonas aeruginosa IA7d terhadap kapang fitopatogen Fusarium solanii dan Pythium sp.. Penelitian ini menggunakan rancangan acak lengkap dengan 16 perlakuan dan 3 kali ulangan pada setiap perlakuan. Biosurfaktan diujikan pada beberapa konsentrasi (0, 100, 500, 1.000. 5.000, 10.000, dan 20.000) ppm, serta larutan Tween-80 20.000 ppm sebagai kontrol positif. Kemampuan penghambatan pertumbuhan dan nilai MIC biosurfaktan diukur dengan metode difusi dan dilusi. Data diameter daerah penghambatan dianalisis secara statistik dengan Kruskal-Wallis test dan uji Dunnett'sT3 Sedangkan data dilusi dianalisis secara deskriptif Hasil penelitian menunjukkan bahwa biosurfaktan Pseudomonas aeruginosa IA7d menghambat pertumbuhan Fusarium solanii dan Pythium .sp. Perbedaan konsentrasi biosurfaktan Pseudomonas aeruginosa IA7d berpengaruh terhadap diameter daerah penghambatan pertumbuhan Fusarium solanii dan Pythium sp.. Penghambatan terbesar dari biosurfaktan Pseudomonas aeruginosa IA7d terdapat pada Fusarium solanii dengan diameter daerah penghambatan sebesar (10,257 ± 1,068) mm pada konsentrasi 20.000 ppm. Penghambatan terkecil dari biosurfaktan Pseudomonas aeruginosa IA7d terdapat pada Pythium sp. dengan diameter daerah penghambatan sebesar (1,637 ± 0,153) mm pada konsentrasi 100 ppm. Nilai MIC biosurfaktan Pseudomonas aeruginosa IA7d terhadap Fusarium solanii dan Pythium sp. adalah sebesar 100 ppm. Translation: Biosurfactant of Pseudomonas aeruginosa IA7d had an ability to inhibit microbial growth. This research was done to know the inhibition potential and MIC (Minimal Inhibitory Concentration) value of Pseudomonas aeruginosa IA7d biosurfactant against phytophatogenic fungi Fusarium solanii and Pythium sp.. This research used randomized complete design with 16 treatments and 3 replication for every treatments. Biosurfaktan was tested in different concentrations (0, 100, 500, 1.000, 5.000, 10.000, and 20.000) ppm, and Tween-80 20.000 ppm as positif control. Inhibition potential test and MIC value were measured using diffusion and dilution methods. The data of inhibition zone diameters was analyzed statisticly using Kruskal-Wallis test and Dunnet s T3 test. The dilution data was analyzed descriptively. The result showed that Pseudomonas aeruginosa IA7d biosurfactant could inhibit Fusarium solanii and Pythium sp. growth. Different concentration of Pseudomonas aeruginosa IA7d biosurfactant influenced inhibition zone of Fusarium solanii and Pythium sp. growth. The biggest inhibition of Pseudomonas aeruginosa biosurfactant was Fusarium solanii at 20.000 ppm with inhibition zone diameter about (10,257 ± 1,068) mm. The smallest inhibition of Pseudomonas aeruginosa IA7d biosurfactant was Pythium sp. at 100 ppm with inhibition zone diameter about (1,637 ± 0,153) mm. MIC value of Pseudomonas aeruginosa IA7d biosurfactant against Fusarium solanii and Pythium sp. were 100 ppm.
