Abstract
Penelitian ini bertujuan untuk mengetahui pengaruh polisakarida krestin (PSK) Coriolus versicolor dengan pemberian waktu berbeda terhadap konsentrasi IL-10 dan histologis paru mencit yang terpapar Mycobacterium tuberculosis. Tiga puluh mencit betina strain Balb/C, berumur 8-10 minggu dibagi menjadi 6 kelompok perlakuan, sebagai berikut: Kelompok K sebagai kontrol normal, K+ sebagai kelompok kontrol positif dengan pemberian PSK saja, K- sebagai kelompok kontrol negatif dengan paparan Mycobacterium tuberculosis saja, P1 dengan pemberian PSK selama 7 hari sebelum pemaparan M. tuberculosis, P2 dengan pemberian PSK selama 7 hari setelah pemaparan M. tuberculosis dan P3 dengan pemberian PSK selama 7 hari sebelum dan sesudah pemaparan Mycobacterium tuberculosis. Polisakarida krestin 0,2 mL diberikan melalui gavage dengan dosis 50 mg/kg dan pemaparan M. tuberculosis dilakukan melalui intraperitonial. Indikator respons imun mencit yaitu konsentrasi IL-10 yang diukur dengan ELISA serta histologis paru mencit dengan metode parafin dan pewarnaan Hematoxylin-Eosin. Data konsentrasi IL-10 dianalisis Brown-Forsythe dilanjutkan uji t sedangkan histologis paru dilakukan skoring kemudian dianalisis dengan Kruskall Wallis. Data hasil pemberian PSK menurunkan konsentrasi IL-10 pada kelompok perlakuan P3 sebesar 21,85 pg/mL tetapi tidak berpengaruh terhadap histologis kerusakan paru akibat paparan M. tuberculosis.
