Abstract

The aim of this research is to synthesize O-(3’,4’- dichlorobenzoyl)-5-nitrosalicylic as an aspirin derivate and to examine its analgesic activity in mice. O-(3’,4’-dichlorobenzoyl)-5-nitrosalicylic was synthesized with esterification method by reacting 5-nitrosalicylic acid with 3,4-dichlorobenzoyl chloride. Tetrahydrofuran (THF) and pyridin was used as a solvent and respective catalyst. The result obtained for the synthesize of 60% in the form of yellowish/tawny powder. The resulted compound was recrystallized by using ethanol and its purity was tested by thin layer chromatography and melting point test. The structure was confirmed by UV spectrophotometry method, infrared spectrophotometry method, and HNMR spectrometry method. Its analgesic activity was tested with Writhing test method. The percentages of pain-inhibition for O-(3’,4’-dichlorobenzoyl)-5- nitrosalicylic for the dose 25mg/kg body-weight was 38,68%, for 50 mg/kg body-weight was 68,68% and for 100 mg/kg body-weight was 78,79%. On the other hand, the percentages of pain-inhibition for the standard compound aspirin for the dose 25mg/kg body-weight was 32,42%, for 50 mg/kg body-weight was 60,99% and for 100 mg/kg bodyweight was 76,92%. In conclusion, the aspirin derivate that was synthesized which is O-(3’,4’-dichlorobenzoyl)-5-nitrosalicylic have higher analgesic activity than aspirin.
