Abstract

The derivate of salicylic acid was designed and synthesized. The objective of this research is to synthesize 2-(3ꞌ,4ꞌ- dichlorobenzoyloxy)benzoate acid, derived from salicylic acid and to measure its analgesic activity in mice (Mus Musculus). The compound was synthesized by reacting salicylic acid and 3,4- dichlorobenzoyl chloride in the presence of pyridine. This reaction was synthesized using esterification method and pyridine was used as catalyst and tetrahydrofuran (THF) as solvent. The synthesized product was recrystalilized by using methanol and its purity was tested by using thin layer chromatography and melting point test. Then, it was analyzed using UV-Vis spectroscopy, infrared spectroscopy, and 1H-NMR spectroscopy method. The percentage of the synthesized product using these reagent and preparation method are 68,14%. Its analgesic activity was tested with writhing test method. The pain-inhibition percentage of 2-(3ꞌ,4ꞌ- diclorobenzoyloxy)benzoate acid for 25mg/kg body-weight was 30,3%. In the dose 50 mg/kg body-weight was 58,9% and for 100 mg/kg body-weight was 71,1%. However, the standard compound asetosal in the dose 25mg/kg, the pain-inhibition was 22,9%, 50 mg/kg body-weight was 53,5% and 100 mg/kg body-weight was 70,8%. ED50 value of 2-(3ꞌ,4ꞌ-diclorobenzoyloxy)benzoate acid is 44,52 mg/kg and lower than salicylic acid 51,34 mg/kg. In conclusion, 2-(3ꞌ,4ꞌ-diclorobenzoyloxy)benzoate acid has higher analgesic activity than asetosal.
