Abstract

The combination of amoxicillin and clavulanic acid is a widely used as oral combination of antibiotic and a beta-lactamase inhibitor i.e. clavulanate potassium (potassium salt of clavulanic acid). In present study, two spectrophotometric methods, derivative and threewavelength UV spectrophotometry using methanol as solvent, were developed for the determination of amoxicillin and clavulanate potassium in tablet simulation. In first derivative UV spectrophotometry, dA/dλ values were measured at 224 nm for amoxicillin and 256 nm for clavulanate potassium. Calibration graphs were linear over the range 10,0-36,0 mg/l of amoxicillin and 2,5-9,0 mg/l of clavulanate potassium in all two methods. Mean recoveries and the standard deviations of amoxicillin and clavulanate potassium were found as 108,64% ± 4,65 and 119,28% ± 10,05 respectively. In threewavelength UV spectrophotometry, absorbance values were measured at 226 nm, 232 nm, 238 nm for amoxicillin and 262 nm, 268 nm, 274 nm for clavulanate potassium. Mean recoveries and the standard deviations of amoxicillin and clavulanate potassium were found as 107,93% ± 2,94 and 134,71% ± 13,09 respectively. All the results were compared statistically. The method validation data showed a high degree of linearity and a high degree of precision results but a low degree of accuracy.
