Abstract

Background: The number of failures in pulp capping treatment were still high, the cause of damage in teeth was caries. The material that has been used for pulp capping causes tunnel defect and necrosis in pulp so an alternative material was required. Propolis was known to be containing resin and has anti-inflammatory properties which can be used as a material for pulp capping. Several researches have used propolis as a material for pulp capping but the mechanism as to how the propolis stimulated the formation of reparative dentin remained unclear.The objective of this study was : to reveal the molecular mechanism of propolis stimulation on odontoblast like cells exposed to inactive Lactobacillus acidophilus in inducing differentiation of pulp fibroblast. Methods: this research was done in pulp cell culture induced by inactive Lactobacillus acidophilus and exposed to propolis extract. Pulp cell culture isolated from human impacted thirds molar that has been extracted. Observation and measurement of the expression of TLR2, NF #954; , TNF , TGF- 1, TGF- R, collagen type 1, DMP-1 was done, also the observation of ALP activity was done by using immunocytochemistry technique and colorimetric to verify the effect of propolis exposing to the differentiation of pulp fibroblast. Result: Data analysis with Manova test and regression test, a significant difference in every group (p<0.05) was present, and showed that there is causalistic relationship among TLR2, NF #954; , TNF , TGF- 1, TGF- R, collagen type 1, DMP-1, and ALP activity. Conclusion: molecular mechanism of propolis stimulation on odontoblast like cells exposed to inactive Lactobacillus acidophilus in inducing differentiation of pulp tissue fibroblast through the inhibition of TLR2, NF #954; , TNF expression and enhancement of TGF- 1, TGF- R, collagen type 1, DMP-1 expression and ALP activity.
