Abstract

The present study was designed to investigate the effect of several factors in corn cob culture medium on growth and red biopigment yielded by Penicillium local Baluran. Wild fungal was isolated from soil at Baluran National Park, Indonesia and identified as Penicillium resticulosum Blr1. Biomass and red bio-pigment yield P.resticulosum Blr1 were affected by type of carbon, combination of carbon and nitrogen concentration, initial pH, media formulation and incubation period in shaken cultures. Cellulose and xylose are the best carbon sources for growth and red bio-pigment yield of P.resticulosum Blr1 in hydrolisat culture media. On the other hand, no differences biomass and biopigment yields P.resticulosum Blr1 in NaNO3, NH4NO3, yeast extract and peptone in corn cob hydrolysate culture medium. Microcycle conidiation of P.resticulosum was attained in three shaken cultures under initial pH 5.5, at 28-30oC with 60-70% humidity relative, at agitation 60 rpm for 12 days incubation period in dark condition. Penicillium isolate local Baluran was identified as P.resticulosum Blr1 can produce the red biopigmen at conidiating, The highest biomass growth 5.35 ± 0.30 g/l and P. resticullosum Blr1 red biopigment yield (0.636 ± 0.075 g/l) were obtained in complex culture medium compose of corn cob (50.0 g/l), yeast extract (3.0 g/l), ammonium nitrate (4.0 g/l), sodium nitrate (3.0 g/l), KH2PO4 (5.0 mg/l), MgSO4.7H2O (0.02 g/l) and water (1000 ml). This study indicated that P. resticullosum Blr1 has hight potential as novel candidate red biopigment producer. Complex culture medium compose of corn cob (50.0 g/l), yeast extract (3.0 g/l), ammonium nitrate (4.0 g/l), sodium nitrate (3.0 g/l), KH2PO4 (5.0 mg/l), MgSO4.7H2O (0.02 g/l) and water (1000 ml) under initial pH 5.5 is a new culture media for red biopigment production.
