Abstract

The purpose of this study was to determine the character of antigenicity Outer Membrane Protein of local isolates Brucella abortus that can be used as a candidate diagnostic kitt. The samples of bacteria were isolated from infected cattle with B. abortus in South Sulawesi and East Nusa Tenggara. They were characterized by conventional test and genomic PCR. For knowing the homology level and phylogenetic relationship, nucleotides seguence of omp2a gene fragment of local isolate B. abortus were compared to isolate from other countries that were accessed in GenBank. Characterization of protein profile local isolate B. abortus based on molecular weight was determined by Sodium Dodecyl Sulphate – Polyacrylamide Gel Electrophoresis (SDS-PAGE), while characterization of protein antigenic profile was examined by Western Blotting. The reactivity of antigenic OMP were examined against bovine brucellosis sera by indirect Enzyme Linked Immunosorbent Assay (ELISA) method. The results showed that the OMP B. abortus local isolates compared with B. abortus isolates from another country have a high homology level between 99% to 100%. The results of protein separation by SDS-PAGE method and Western blot showed that the OMP with a molecular weight of 33 kDa was antigenic. The results of indirect ELISA showed that optical density (OD) value of OMP B.abortus local isolate from South Sulawesi was higher than isolate from East Nusa Tenggara, so this OMP can be used as a candidate diagnostic kitt.
