Abstract

The objective of this study is to determine the effect of crude extracts Phyllophorus sp to phagocytic activity, levels of IL-12, and the diameter of the germinal center spleen, also to identify extract with the best potentially immunomodulatory. Thirty-six male mice divided into two large groups of 1 and 2, then put into 2 sub-group, which included two control groups were given 0.5% CMC (negative control) and 0.5% CMC and bacterial infection (positive control), and 4 treatment groups were fed crude extract, n-hexane extract, ethyl acetate extract and final ethano extract. First fourteen days, all groups were treated extracts, except for negative control group and positive controls, with a dose of 0.0462 g of dry weight of sea cucumbers per day. Day 15th all treatment groups except the negative control was injected intraperitoneally M. tuberculosis 106 and be repeated injection in group 2 on the 18th day. For group 1 mice were sacrificed on 17th day and intraperitoneal fluid collection conducted to test the activity of phagocytosis and blood for measurement of serum levels of IL-12.Group 2 dissected on day 28th and harvesting spleen for measuring the diameter of the germinal center spleen by the paraffin method and Hematoxylin-Eosin staining. The data obtained and analyzed one-way Anova ( = 0.05), followed by Duncan test. From the results of treatment of crude extracts affect the levels of IL-12 and diameter of the germinal center spleen but not to the phagocytic activity. Extract that have the potential as an immunomodulatory is the n-hexane extract because it has levels of IL-12 of the most high and the largest of the diameter spleen germinal centers.
