Abstract
This research is to analyze the nifedipine effect on the decrease of epithelium thickness, the increase in seminiferous tubules lumen diameter, and expression decrease of the mouse sperm membrane protein profile. This study used 24 strains of mice BALB / C adult weight 20-30 g, divided into four groups. Each group consisted of six rats. The first group was a control group given 0.5% CMC solution 0.01 ml / g / bw / day orally, whereas treatment groups were given different doses of nifedipine successively 3.75; 7.5; and 15 #956;g /g body weight /day orally. Measured parameters were the thickness of the epithelium, lumen diameter of the seminiferous tubules and sperm membrane protein profile of mouse. Data were tested with one-way ANOVA, and to know the differences among the treatments, it was followed by a least significant difference test with significance level of 0.05. Results showed that, the greater dose of nifedipine, the epithelium thickness of seminiferous tubules decreased successively 57.12, 53.27, and 50.07 #956;m compared to the control group (58.26 #956;m). This result is inversely proportional to the diameter of the seminiferous tubules lumen that are in fact the greater the dose of nifedipine, the diameter of the seminiferous tubules lumen increased respectively 38.46, 42.02, and 45.87 #956;m compared to the control group (36.54 #956;m). From the electrophoresis results showed that there were different protein profiles between control and nifedipine treated groups. The 58 and 38 kDa protein band appeared in the control group and in the treatment dose nifedipine 3.75 and 7.5 #956;g /g bw, but it did not appear in the treatment group nifedipine 15 #956;g/g body weight. The conclusions of this research are 1) Provision of nifedipine reduces the thickness of the epithelium, and increases the diameter of the lumen of the seminiferous tubules of mouse. 2) The provision of nifedipine causes 58 and 38 kDa protein was expressed minimally.
