Abstract
ISOLATION AND CHARACTERIZATION OF CHONDROITIN SULFATE FROM CHICKEN SHANK AS THE RESULTS OF FARM WASTE Rizky Amalia Adlina Affandhi This work was aimed to study the preparation and hydrolyzation of cartilage from chicken shank to obtain crude chondrotin sulfate (CS). Chondroitin sulfate is used extensively as a treatment for osteoarthritis. This study was conducted to evaluate whether chondroitin sulfate could be isolated from chicken shank cartilage in sufficient quantities and of requisite quality to make it a feasible source of chondroitin sulfate. To obtain CS from cartilage part of chicken shank, use of enzyme (papain 50 g/ 550 g of cartilage), acid (acetic acid), and heat treatment were studied. The cartilage obtained from chicken shank was hydrolysed by papain for 24 h, extracted by incubation at pH 4.5 and 37o C for 6 h and 8 h, and the supernatan was treatment by cetylpyridinium chloride (CPC), NaCl, methanol and ethanol. The precipitate obtained from extraction was dried in oven at 55o C. The crude CS obtained was identified using Fourier Transform – Infrared (FT-IR) and spectrophotometric method. Crude CS obtained from 6 h of incubation was 16,30% and from 8 h of incubation was 36,35 % (dry weight). The FTIR spectra of CS from shank cartilage was identical with CS standart. Quantitative analysis on this study uses spectrophotometric. The precision was good with RSD< 7,3% and the accuration was good with %recovery 82,48-108,88%. This study shows that chicken shank cartilage is a readily available source of chondroitin sulfate.
