Abstract

Osteoporosis is a disease characterized by low bone mass and structural deterioration of bone tissue. Estrogen deficiency causes loss of bone mineral density which affects osteoporosis. Phytoestrogen was an potential alternative of estrogen that has minimum side effects. Many edible plants contain of phytoestrogens that are believed to promote bone health. Abelmoschus manihot (L.) Medik is one of the plants in Indonesia which suspected contains estrogen like substance and potential to prevent osteoporosis. The aim of this research is to determine whether the Abelmoschus manihot (L.) Medik leaves has the potential to increase bone formation at in vitro assays in preosteoblast cell line MC3T3-E1. Abelmoschus manihot (L.) leaves is extracted with ethanol 96 %. Identified with GC-MS and tested through in silico study in ERβ to know it’s affinity, and compared it to estrogens affinity. The results of in vitro study showed that phytoestrogen from ethanol 96 % extract is able to increase bone formation by increasing ALP activity that is known from CLSM. Through correlation analysis of in vitro results, TIC GC-MS, and in silico results.
