Abstract

Andrographolide is a major component from plant Andrographis paniculata which has wide pharmacological activities, however this substance has been limited by poor solubility, short half-life, low bioavailability and instability in extreme acidic or alkaline conditions of gastrointestinal tract. A method was used to overcome these problems which was microparticle systems from matrix chitosan and prepared by ionic gelation-freeze drying method using tripolyphosphate as the cross linker. In this study, microparticles were prepared with different type and concentration of chitosan. Microparticles formed as yellowish white powder, slightly bitter and have no odor. All formulas have spherical shape and heterogenous in size. Infrared spectra and DTA thermogram indicate ionic bonding between chitosan and TPP. XRD analysis exhibit its crystallographic structure and verify that the crystallinity of andrographolide has changed. All formulas were easy to redisperse in aquades. The results from the entrapment efficiency evaluation show that concentration increasing, significantly decrease the microparticles entrapment efficiency while the two types of chitosan used have no impact in microparticles entrapment efficiency.
