Abstract

Cyprinid herpesvirus 3 (CyHV-3), the etiological agent of Koi Herpesvirus Disease (KHVD,) is an emerging agent that causes fatal disease in koi. Since its first emergence in Blitar, it has spread to most regions in Indonesia due to fish trade and ornamental koi shows that has caused severe financial and social losses. This research aims to determine molecular characterization of glycoprotein encoding gene (ORF25) from CyHV-3 through analysis of nucleotide sequences and amino acid also to predict the epitope of isolate CyHV-3 in East Java. The results showed that isolate from East Java is related closely to isolates from Japan, China and Korea. The nucleotide sequences showed the highest sequence similarity up to 99.7% to isolate from Japan. The results from epitope prediction from T cell in ORF25 showed that isolate from Malang has the lowest epitope cell while isolates from Madiun, Kediri and Blitar has each 39 epitope cells. Epitopes B-cell based Bepipred Linear Epitope on ORF25 protein isolate Malang states have candidates 33 B-cell epitope, while at Madiun and Blitar isolate has 31 candidates B-cell epitopes These findings indicated that isolate of CyHV-3 from East Java can be used as a vaccine candidate or diagnostic kit for early detection of KHVD appropriately.
