Abstract

Embryo Transfer was one ofreproduction technology used to increase cattle productivity. The succeed ofembryo transfer depends on the quality ofthe embryo. The Quality ofoocytefor in vitrofertilization in order to produce in vitro embryo was influenced by the in vitro maturation process. In order to reveal the maximum competence, the growth of the oocyte was also influenced by somefactors produced by thefollicle which has thefunction to initiate growth into primaryfollicle. One of thosefactors was Kit Ligand. Kit Ligand (KL) was a growthfactor bind to C-Kit receptor which was actively involved in initiating the beginning ofprimordialfollicle growth process in the ovary. The lack presence of Kit Ligand could cause malfunction in the oocyte growth. "lhs research consisted of collecting oocyte and in vitro culturing, protein isolation, electro elution to purified Kit-Ligand isolate, producing polyclonal antibody, immunization on experimental animal, and Specification test ofKit Ligand using dot blotting method. The presence Ofcolor changing showed specification between thé protein as antigen and the polyclonal antibody. Dot blotting method could also confirm the concentration of antibody based on the time difference on collecting sample. The result showed on the second week the immune respond increased. It increased gradually until the fourth week (second bleeding) and then decreased. The production of antibody began to increase again and reach the top on the eighth week (sixth bleeding).
