Abstract

Newcastle Disease is a highly contagious and an endemic disease in Indonesia. Vaccination program has failed to bring protection. This is because of the distant similarity of vaccine strains and local strains; vaccine strains belong to class II genotype II while local NDV strains belong to class II genotype I, VI, and VII. Therefore, there is a need of local NDV genomic data recording to provide more information for better protection. Ten lentogenic NDV isolated from domestic swan geese (Anser cygnoides domesticus) in Lumajang and Surabaya, East Java, Indonesia were analyzed for its homology with other isolates circulating in Indonesia according to its HN protein. Death times of these samples were observed in 9 old days SAN embryonated eggs. Four (Ags/K3, Ags/K4, Ags/L8, Ags/L9) out of ten samples showed death time more than 90 hpi, high HA titres, and HI titre of >26. Partial HN genes of 503bp of samples were then amplified using One Step RT-PCR and visualized using agar gel electrophoresis. Amplified products were then purified and sequence were analyzed. Protein sequences were then aligned using ClustalW Alignment in BioEdit version 7. Homology scores between amplified amino acid sequence of each samples and reference sequence were then analyzed using Needleman-Wunsch Algorithm via NCBI, they showed low similarity of 18% - 43%.
