Abstract

Aim of this research was to know how much dilution of cat blood that can still be detected by Leucomalachite Green (LMG) and Takayama reagent. This research was a laboratory exploratory study. Three samples of domestic cat blood were diluted with ratio 1:10; 1:100; 1:1,000; 1:10,000; and 1:100,000. Bloodstain is made by dropping each blood dilution on the filter paper for LMG test and on the object glass for Takayama test. Bloodstain for LMG test was done with ten repetitions from each samples and duplo for Takayama test. Positive results from the LMG test are presented as a bluish-green discoloration in stains. A positive result from the Takayama test is the formation of hemochromogen crystals under a microscope with 400x magnification. The color change of LMG test reaction are captured and processed with NIH ImageJ version 1.52a software to know the RGB (Red, Green, Blue) value. The value then processed using ANOVA. The dilution of 1:10,000 was detected LMG with the average RGB value 253.970 ± 0.418. While Takayama test can only detected sample of 1:1,000 dilution. Based on LMG test result in the first series sample dilution, the higher dilution was made between the ratio of 1:10,000 and 1:100,000. The second series of sample dilution were made with ratio 1:20,000; 1:40,000; and 1:80,000. The second series then tested for LMG test with ten repetitions from each dilution. The result of LMG test that can still detected positive was 1: 40,000 dilution with the average RGB value 254.946 ± 0.054
