Abstract

The aim of this study was to examine the antibacterial activity of the methanolic extracts of Callyspongia sp., Niphates olemda, Coelocarteria agglomerans, and Cinachyrella sp. collected from Tabuhan Island, Banyuwangi. The antibacterial activity was undertaken by using microdilution method against Bacillus subtilis ATCC 6633 and Pseudomonas aeruginosa ATCC 27853. A coloring reagent p-iodonitrotetrazolium chloride (INT) was added to the solution to indicate the present of living bacteria. The Minimum Inhibitory Concentration (MIC) was determined as the lowest concentration of samples with no apparant growth of bacteria indicated by no visual changes in INT colour. The results showed that the methanolic extract of Callyspongia sp. has an antibacterial activity against Bacillus subtilis and Pseudomonas aeruginosa. The Minimum Inhibitory Concentration (MIC) inhibited the growth of Bacillus subtilis and Pseudomonas aeruginosa at 500 ppm, whereas the Minimum Bactericidal Concentration (MBC) at 500 ppm. The phytochemical screening of the methanolic extract of Callyspongia sp. indicated that the sample contain alkaloid and terpenoid. The conclusion of this research was among the sponge, only methanolic extract of Callyspongia sp. showed the medium an antibacterial activity against Bacillus subtilis and Pseudomonas aeruginosa.
