Abstract

Paracetamol (acetaminophen) is an active metabolite of phenacetin with antipyretic and analgesic properties. Orally disintegrating tablets of paracetamol were prepared by freeze drying method using mannitol based excipient. This study aims to determine the characterization of paracetamol orally disintegrating tablet (ODT) using polyplasdone XL-10 5% and 10% as disintegrant and gelatin 2% as binder. Freeze dried ODT, physical mixture, and single compounds were characterized using powder X-ray diffractometry (PXRD), differential thermal analysis (DTA), and Scanning Electron Microscope (SEM). PXRD study indicated that there was a decrease in degree of crystallinity of paracetamol in freeze dried ODT compared to physical mixture. DTA study showed that there was a decrease of melting point in the freeze dried ODT but the enthalpy of paracetamol could not be determined because overlapping of endothermic peaks occurred between paracetamol and other excipients. The lowest degree of crystallinity of paracetamol and melting point was showed by formulation containing polyplasdone XL-10 10%. The characterization using SEM showed that tablets prepared by freeze drying method were porous, even though the porosity among all formulations could not be directly determined visually
