Abstract

The aim of this study was to observe the characteristic of dosage form and penetration of diclofenac sodium with microemulsion w/o system in HPC M gel base. The ratio of surfactant (Span 80-Tween 80): cosurfactant (Ethanol 96%) = 6:1 was used to make microemulsion. Diclofenac sodium gel with emulsion was used as comparator. The evaluation included organoleptic, pH, spread diameter measurement of zero load, and also diclofenac sodium penetration test from gel base of each formula. The result of pH test, spread diameter measurement of zero load, and flux from drug release test were evaluated based on independent sample t-test. The result showed that microemulsion system in gel base (formula I) had thicker consistency than emulsion in gel base (formula II). Microemulsion in gel base didn’t show significant difference on pH compared to emulsion in gel base, but showed significant difference on spread diameter measurement of zero load and flux of diclofenac sodium. Data analysis showed that pH for formula I was 6.61 ± 0.05 and formula II was 6.70 ± 0.06. Spread diameter measurement of zero load for formula I was 6.63 ± 0.22 cm and for formula II was 10.45 ± 0.05 cm. Drug penetration test was carried out with Erweka Dissolution Tester Type DT 820 with apparatus 5 paddle overdisk in phosphate buffer 7.4±0.05, temperature 37°C, 100 rpm. The rate of diclofenac sodium release in formula I was 0.4168 ± 0.0483 μg/cm2/menit. Formula II product couldn’t be made because its unstable.
