Abstract

Paracetamol is usually given orally as antipiretic and analgesic for the treatment of fever and the symptoms associated with fever. Pediatrics may have difficulty ingesting conventional tablet because of their underdeveloped muscular and nervous systems. This problem has been solved by oral dosage form which can disintegrate rapidly and dissolve in mouth within less than 1 minute without the need for water, namely orally disintegrating tablet (ODT). This study aims to determine the characterization of paracetamol ODT using mannitol as filler, gelatin as binder, and polyplasdone XL-10 as disintegrant prepared by freeze drying method. Paracetamol ODT, physical mixture, and single compound of the material were characterized using Xray Diffractometry (XRD), Differential Thermal Analyzer (DTA), and Scanning Electron Microscope (SEM). XRD study demonstrated that there was significant decrease of paracetamol’s intensity in freeze dryed ODT compared to the physical mixture in the formula with gelatin 1% (F2) and gelatin 2% (F3) at 2θ 12,1o and 13,7o, but there was no significant decrease between the freeze dryed ODT F2 and F3. DTA study showed that paracetamol enthalpy could not be determined because overlapping of endothermic peaks occured between paracetamol and other excipients. The characterization using SEM showed that paracetamol ODT prepared by freeze drying method was porous.
