Abstract

Avian influenza (AI) H9N2 virus has been identified in domestic birds and wild birds in various parts of the world and has become one of the dominant AI virus subtypes. The purpose of this study was to evaluate H9N2 virus infection in apparently healthy poultry in Sepanjang live-poultry market. A total of 135 chicken and duck cloacal swabs were collected from Sepanjang live-poultry market and inoculated on embryonated chicken egg. There were 21 positive samples by HA test and two samples were positive for H9 avian influenza virus using Real Time RT-PCR test. One positive sample (AN 4-4) was sequenced using Next Generation Sequencing (NGS), the DNA sequences were identical to the H9N2 AI virus reference. Its HA had 98.56% homology to A/chicken/Vietnam/H7F-LC4-51/2014 (H9N2), and its NA had 98.43% to A/chicken/Vietnam/H7F-LC4-371/2014 (H9N2). The amino acid analysis on the cleavage site position showed PSRSSR ↓ GLF that categorized as Low Pathogenic Avian Influenza (LPAI). The receptor binding site had L (leucine) and G (glycine) in the 226 and 228 positions which were similar to mammalian sialic acid (sialic-α2,6-gal), that might give a potential risk to transmitted to humans.
