Abstract

The present study was designed to investigat e decreasing of necrotic and apoptotic muscularis cells and hepar cells in stre ptozotocin-induced diabetic mice treated by vanadyl sulphate. Muscularis and hepar tissue were analyzed with histochemistry using hematoxylin-eosin staning and immunohist ochemistry using Apo-BrdU-IHCTM DNA Fragmentation Assay Kit and caspase-3 stanin g. 25 mice divided into five groups i.e. placebo group, positive group, and three treat ment groups based on dose (5, 30, 100 mg/kg BW). Diabetic mice model were induc ed by twice intraperito neal injection of streptozotocin on day 1 (single dose 10 mg /kg BW) and day 14 (single dose 50 mg/kg BW. Diabetic occurred on day 21 and show n by the significantly increased of blood glucose level from 111.00 ± 6.79 mg/dl to 202.93 ± 12.99 mg/dl. Vanadyl sulphate was given orally to the three treatment groups, once daily from day 21 to 27. Vanadyl sulphate dose 5 mg/kg BW decreased blood glucose level from 163.00 ± 3.06 mg/dl to 102.67 ± 19.03 mg/dl; dose 30 mg/kg BW decr eased blood glucose level from 219.75 ± 22.02 mg/dl to 111.25 ± 19.69 mg/dl and dose 100 mg/kg BW decreased blood glucose level from 242.67 ± 24.55 mg/dl to 62.67 ± 17.65 mg/dl. Results showed that three doses treatment of vanadyl su lphate were significantly reduced blood glucose level compared to positive group (F(4,18) = 6,324 ; p < 0,05). Mor phological change of muscularis and hepar tissues were analyzed by histochemi stry using hematoxylin-eosin staining. The results showed that vanadyl sulphate re stored atrophic and necrotic condition in muscularis to normal and also stimulated he patocytes regeneration in hepar tissue. The qualitative immunohistochemical study’s result showed that apoptotic muscularis and hepar cells decreased in diabetic mice.
