Abstract

This research was conducted to determine the antibacterial effect of Dayak onions to Escherichia coli using dilution method. Dayak onions (Eleutherine americana Merr.) have several active substance as an antibacterial agents such as flavonoids, tanins, terpenoids, alkaloids and phenols. Dayak onions were extracted by maceration method using ethanol 96% solvent. Dayak onions was diluted using aquadest and CMC Na 1%. Escherichia coli is the bacteria that causes the most diarrhea in humans and animals. This research used two type of E. coli, E.coli ATCC 25922 and E. coli environmental isolates. The dilution method used four treatments of each bacteria and two control to determine the Minimum Inhibitory Concentration (MIC) and continued by streaking on the Mueller Hinton Agar (MHA) medium to determine the Minimum Bactericidal Concentration (MBC). The treatment used were 1%, 5%, 10%, 15%, control negative and control positive. The treatment group contained of E. coli and dayak onion ectract, control negative contained only suspention of E. coli, control positive contained only dayak onion extract, each treatment was repeated by three repetition respectively. The MIC value can not be determined because all the MIC test tube show similar turbidity, and there is no turbidity difference between before incubation and the last incubated. MBC test showed different result betwen E. coli ATCC 25922 and E. coli environmental isolates. Minimum Bactericidal Concentration test result showed dayak onion extract treated bactericide to E. coli ATCC 25922 at 5% concentration and MBC test result showed dayak onion extract terated bactericide to E. coli environmental isolates at 1 % concentration.
