Abstract

Background: Regeneration of damaged bones can be done using a replacement material, namely bone graft. Bone graft is used as a filler and scaffold to facilitate bone formation and promote wound healing. There are 4 types of bone graft, one of which is xenograft. Xenograft is bone tissue from another species that has been proven to be used successfully as a bone filling material that is damaged by removing the organic tissue that is inside. Xenograft has various types, including DFDBBX and FDBBX. The combination of cells and growth factors, proven effective for the process of healing bones. One of the growth factors needed for bone regeneration is BMP-4. BMP-4 has a function to induce osteogenic differentiation and increase bone formation. Purpose: To find out the comparison of BMP-4 content in DFDBBX and FDBBX. Methods: In both samples, DFDBBX and FDBBX, protein extraction was carried out using the guanidine HCl method and centrifugation was performed to obtain supernatant. The supernatant is inserted into the plate and replicated 5 times on each sample to be tested for protein content using ELISA. The absorbance is read using an ELISA reader with a wavelength of 450 nm. The absorbance value is then converted using the formula to get BMP-4 content in DFDBBX and FDBBX (ng/mL). Data analysis using Mann Whitney Test. Results: From these two samples, DFDBBX and FDBBX, it shows that the graft contained BMP-4 growth factor. The average yield of BMP-4 content in DFDBBX is 2.6158 ng/ml and FDBBX is 3.1686 ng/ml. Test results Mann Whitney show p=9,17. Conclusion: There are differences in BMP-4 content between DFDBBX and FDBBX but unsignificantly.
