Abstract
Latar Belakang: Ulkus traumatik adalah lesi yang disebabkan oleh peningkatan Advanced Glycosylation End Products (AGEs). AGEs meningkatkan Reactive Oxygen Species (ROS) intraseluler, sehingga meningkatkan ekspresi NFқB dand COX-2 yang dapat memperpanjang waktu penyembuhan ulkus. Gel propolis memiliki efek antiinflamasi yang dapat mempersingkat waktu penyembuhan ulkus. Obyektif: Menganalisis potensi gel propolis dalam menurunkan ekspresi NFқB and the COX-2 pada ulkus traumatik tikus Wistar (Rattus novergicus) diabetik. Metode: Penelitian menggunakan posttest-only control groups. Sebanyak 27 ekor tikus Wistar jantan dibagi rata dalam 3 kelompok: kelompok pertama, tikus Wistar nondiabetik; kelompok kedua, tikus Wistar diabetik diaplikasikan dengan gel HPMC 5%; and Kelompok ketiga, tikus Wistar diabetik diaplikasikan dengan propolis gel. Ekpresi NFқB dan COX-2 diamati pada hari ke-1, ke-3, dan ke-5 dengan pemeriksaan Histologi dan Immunohistokimia menggunakan antibodi monoklonal NFқB (Santa Cruz Biotechnology, Inc, sc-8008) dan antibodi monoklonal COX-2 (Santa Cruz Biotechnology, Inc, sc-1747). Hasil: Gel propolis menurunkan ekspresi NFқB (p= 0.002 and p= 0.000) pada kelompok perlakuan hari ke-3 dan ke-5. Menurunkan ekspresi COX-2 (p= 0.003) pada kelompok perlakuan hari ke-5. Kesimpulan: Aplikasi Topikal gel propolis dapat meningkatkan penyembuhan ulkus traumatik diabetik pada model tikus yang diinduksi Diabetis Mellitus Tipe I dengan injeksi Streptozotocin.
