Abstract

Tablet manufacturing process can induce mechanical and thermal energy forming an eutectic. Ibuprofen (IBU) and stearic acid (AST) are known interact to form an eutectic. Aim of this research is to determine effect of compression force on physical characteristic of IBU-AST binary mixture. IBU:AST with ratio 40:60, 50:50, and 60:40 (w/w) was compressed under various compression force using hydraulic press plain flat punches diameter of 13 mm. Physical characterization was performed 2 days to allow particle recovery, then tablet hardness properties was tested. Physical characterization of this mixture due to compression force has been studied using differential thermal analysis (DTA), X-ray powder diffraction (XRPD), fourrier transform infrared spectroscopy (FT-IR), and scanning electron microscope (SEM). According to the hardness test result, ratio of 40:60 have the highest tensile strength on 170,6 N/cm2 with 19,93 kN compression force, higher than other composition and its single entity. Thermal analysis DTA showed that ratio of 40:60 is the eutectic composition with single endotermic peak at 53,2 °C, lower than other composition and its single entity. XRPD study showed diffraction peak shift of this composition at the compression force of 19,93 kN on 2θ = 6,6°; 8,8°; and 11,2°, with the decreased intensity on higher compression force. Scanning electron micrograph showed sintering phenomenon. New transmission peak at 1448 cm-1 indicating asimetrycal bending vibration of C-H from metyl and changing of spectra at 2692 cm-1 on –OH strecthing region showed through FT-IR study. All of these phenomena indicate physical interaction of IBU-AST due to compression force.
