Abstract

The aim of this study was to screening fibrinolytic enzyme producing bacteria from soil samples collected from Mangrove Ecotourism Beach Wonorejo, Surabaya. The isolation of samples were done by serial dilution and plating methods. Qualitative test to the bacteria that produce protease enzyme was conducted by using skim milk agar medium. Bacteria that have proteolytic activity were characterized by presence of clear zone around of the growing bacteria. A total of 16 from 20 bacteria were screened based on the zone of proteolytic. The bacteria that gives proteolytic activity were screened for fibrinolytic activity on fibrin plate. The result of screening, were 12 isolates bacteria from 16 had fibrinolytic activity, with enzyme activity index highest was 1.71 (BM 9.1). All the bacteria that have fibrinolytic activity were characterization by microscopic and macroscopic. From the twelve were isolated bacteria above, two of them with the highest index (BM 8.1 and BM 9.1) were selected to be identified using biochemical identification. The result of identifying, the bacteria was Bacillus sphaericus (BM 8.1) and Bacillus megaterium (BM 9.1)
