Abstract
Cancer treatment often fails due to various reasons such as many cancer cells are not considered as a foreign substance by host cell, cancer cells quickly become resistant to anticancer drugs, and many cancer drugs are toxic. Therefore, a new drug is needed to provide promising candidate as anticancer agent which has higher biological activity and lower toxicity. Folic acid analog is one of new drug that provide promising prospects as anticancer agent which competitively inhibits dihydrofolate reductase. It is necessary to modify folic acid molecule to produce another analog which is expected to provide a specific and optimal activity. Folic acid modification begins with in silico test to predict the activity of compound which showed that 10-N-(4′-bromobenzoyl)folic has higher biological activity than methotrexate as reference. Synthesis of 10-N-(4′-bromobenzoyl)folic acid by reacting folic acid with 4-bromobenzoyl chloride through acylation reaction principle, percentage yield of this synthesis was 13%. The purity of product was analyzed by using thin layer chromatography and the structure of product is confirmed by using 1H-NMR. At last, its cytotoxic activity was determined by using MTT assay method against HeLa cell line compared with methotrexate as reference drug. The result of MTT assay of both compound, 10-N-(4′-bromobenzoyl)folic acid and methotrexate against HeLa cell line did not show any significant cytotoxic activity up to 2000 ppm. It was concluded that 10-N-(4′-bromobenzoyl)folic acid could be synthesized through acylation reaction, and 10-N-(4′-bromobenzoyl)folic acid and methotrexate cytotoxic activity could not be determined by using MTT assay against Hela cell line.
