Abstract

The increasing oxidative stress is considered to be the main mechanism involved in the pathogenesis of endothelial dysfunction and leading to hypertension. Robusta coffee and corn contain ferulic acid which is antioxidants and antihypertensive, so the coffee-corn mixture has the potential to be an antihypertensive treatment. The aim of this study was to analyze the effect of coffee-corn mixture on NO,eNOS, sodium, ACE and F2- isoprostane serum levels and blood pressure on hypertensive rat models. This research was conducted in 2 phases; the first phase in this study was to quantitatively analyze ferulic acid levels of Robusta coffee bean and corn extracts and the second phase analyzed the mechanism of the effects of coffee-corn mixture toward the increase of nitric oxide, eNOS and the decrease of sodium, ACE, F2-isoprostane serum level and blood pressure on hypertensive rat models. The first phase, ferulic acid levels from samples of coffee from two different regions (Argopuro and Kaliwining, Jember District), and yellow corn from Tanjung’s traditional market were analyzed using Thin Layer Chromatography (TLC)-Densitometry method. In the second phase of the study forty male wistar rats divided into five groups : the negative control group; the positive control groups and the treatment group (hypertension group that were given coffee-corn mixture with the dose of 100mg/kgBB with ratio 100%:0%, 75%:25% and 50%:50% through sondase). Coffee-corn mixture given for 15 days. The day after the end of the fifteenth day the parameters were measured. Results : the first phase showed that the ferulic acid levels obtained from Argupuro coffee bean extract that was roasted at 180°C and 200°C were 1.94 ± 0.02 mg/g and 1.42 ± 0.05 mg/g, while the one from Robusta Kaliwining coffee bean extract roasted at 180°C and 200°C were 33.20 ± 0.07 mg/g dan 2.42 ± 0.20 mg/g. The ferulic acid found on yellow corn extract roasted at 180°C was 1.34 ± 0.06 mg/g while at 200°C it was not used because it was damaged. Kaliwining Robusta coffee is used in the second phase of research. The second phase of the study was a decrease in blood pressure on treatment group compared with positive control group. There was no significant difference on nitric oxide serum levels on negative, positive, and the treatment groups. There was a significant difference on serum levels of eNOS, sodium, ACE, and F2-isoprostane in the negative, positive, and treatment groups. Conclusion: Kaliwining Robusta coffee and coffee-corn mixture with a ratio of 50%: 50% plays a role in reducing blood pressure in hypertension through decreasing ACE, sodium, and F2-isoprostane serum levels
