Abstract

Bone is an important organ that is the main component of the skeleton. Bone defect is a serious condition; if not treated properly, may cause delay or bone healing failure. This study aimed to determine the effect of macrophage polarity on the osteoconductivity of BHA-GEL and HA-GEL scaffold in vivo. The in vivo study model was carried out by implanting BHA-GEL and HA-GEL scaffold into the femoral defect of Wistar rats (defect diameter 2.2 mm, scaffold diameter 2 mm). The scaffold's characteristics in the form of morphology, pore size, and Ca/P ratio were examined using SEM-EDX. The compressive strength was measured using mini autograph. Osteoconductivity was examined based on histological results dan radiological images using X-rays, while macrophage polarity using immunohistochemical techniques. Immunohistochemical results showed that the BHA-GEL scaffold did not show signs of chronic inflammation as seen in the HAGEL scaffold group. Radiological data showed that the BHA-GEL scaffold caused acceleration of defect repair, in line with histological data, which showed accelerated migration of osteoclasts, osteoblasts, and osteocytes. The BHA-GEL scaffold has a round particle shape, while the HA-GEL scaffold has an irregular particle shape. The mean pore sizes of BHA-GEL and HA-GEL scaffold were 0.633 ± 0.089 and 0.361 ± 0.036 μm, respectively (p=0.035). The average compressive strength of the BHA-GEL and HA-GEL scaffolds was 61.714 ± 6.163 and 40.766 ± 4.513 MPa (p=0.041). Meanwhile, the Ca/P ratio of BHA-GEL and HA-GEL scaffold was 1.63 ± 0.05 and 1.44 ± 0.05, respectively (p<0.0001). Based on the study present it can be concluded that the BHA-GEL scaffold is osteoconductive and produces a beneficial immune response compared to the HA-GEL scaffold.
