Abstract

This study aimed to identify the presence of MDR ESBL isolated from rectal swab in pig farm, Malang District, East Java Province using Kirby Bauer Disk Diffusion and PCR method. Isolation and identification in terms of bacteria using selective media and IMViC biochemical test were conducted. Antibiotic sensitivity testing using disc diffusion method was performed on 5 types of antibiotics: Ciprofloxacin, Tetracycline, Trimethoprim, Streptomycin, and Aztreonam. Positive isolates which resistant to 3 or more antibiotic disks then were grouped into MDR bacteria. Then, the MDR Escherichia coli isolates with 4 antibiotics resistance and presumptive ESBL (resistant to Aztreonam) proceed with PCR method to identify the blaCTX-M gene. Positive samples containing E. coli found in 107 out of 120 samples (89%). For antibiotic sensitivity testing, found E. coli which were resistant to Ciprofloxacin (4.67%) Tetracycline (84.12%), Trimethoprim (32.71%), Streptomycin (92.53%), dan Aztreonam (0.94 %). Thirty-two isolates were identified as MDR E. coli, which further isolates with 4 antibiotics resistance and presumptive ESBL tested to PCR for blaCTX-M gene identification. Thus, MDR E. coli were positive in 32 isolates (29.91%) and 1 MDR E. coli isolate was positive presumptive ESBL (3.12%).
