Abstract
The ketogenic diet produces ketones in the blood as an energy source that can be used for various purposes such as nonpharmacological therapy for cancer, weight loss, type 2 diabetes mellitus, epilepsy, and status epileptics therapy in children and adults and can even be used to improve performance in competitive athletes. This study aims to analyze the effect of the ketogenic diet and macronutrient composition on TNFα serum levels in Mus musculus induced with benzopyrene. This was a true experimental study with the randomized posttest-only control group design using 36 female mice (Mus musculus) that were divided into six groups. Dependent variables were body weight, breast tumour area, and TNFα levels. The statistical analysis used SPSS software. Result: the ketogenic diet-induced slower weight gain (p=0.042), lower TNF-α levels (p=0.000) but does not affect breast tumour area. After 42 days, there were slower increased of body weight in the ketogenic diet mice because benzopyrene induced neurotoxicity and the administration of ketogenic diet increased the accumulation of visceral fat and plasma leptin. The ketogenic diet decreased TNF-α levels because it has a neuroprotective and anti-inflammatory effect. However, the ketogenic diet has not been able to reduce the breast tumour area on benzopyrene induced Mus musculus. In conclusion, the ketogenic diet lowers TNF-α levels and slowers weight gain but has not been able to reduce the area of benzopyrene-induced breast tumours. The lowest TNF-α levels were found in groups with a fat:protein ratio = 60:30.
