Abstract
Subclinical mastitis is an intramammary infection characterized by excess immune cells (especially neutrophils) that get in the mammary glands without showing the change. Escherichia coli is one of the most prominent cause of mastitis. The sample used in this study was subclinical mastitis milk taken in October until December 2020 from Surabaya City. Amount of samples was 27 samples. The samples were test by California Mastitis Test (CMT) and 23 samples was positive subclinical mastitis. The 23 samples primary isolated to Brillian Green Bile Broth (BGBB) as medium to maximize coliform bacteria. The secondary isolation was Eosine Methylene Blue Agar (EMBA) as selective medium which aims to seperate the Escherichia coli suspected with other coliform bacteria. Identification of Escherichia coli using biochemical tests, namely TSIA Test which is useful for observe sugar fermentation, SIM Test which is useful for observe motility, SCA Test which is useful for observe citrate permease enzyme, and Urea Test which is useful for observe urease enzyme. The results of the research showed that there were 3 positive samples of Escherichia coli from 27 samples of milk and 23 positive samples subclinical mastitis. The percentage of positive samples of Escherichia coli in subclinical mastitis milk was 13.04% while the percentage of postive samples of subclinical mastitis was 85.18%.
